Highly Branched Oligonucleotides

Data showing capillary electrophoresis (CE) traces of a highly branched oligonucleotide containing a 9-mer thymidine core (T9). Left trace is crude oligo showing where the core and forks elute, where F is the first fork, F2 the second, F3 the third, and F4 the forth fork in the 3' to 5' direction. This was verified on the right trace where a portion of the oligonucleotide synthesis was removed at each addition step and subsequently combined to inject on the CE. Increased coupling times (up to 20 min) and increased total amounts of amidites were needed to ensure high yield.
